Spermidine is a natural polyamine that induces autophagy — the recycling program cells use to clear damaged components, and one that fasting also triggers. In a landmark Nature Medicine study, dietary spermidine extended lifespan in mice and protected the aging heart. In a 20-year Italian cohort, people with the highest spermidine intake had markedly lower mortality — a gap the authors compared to being roughly five to six years younger. Human trial evidence is still early, but the food sources are unambiguously healthy.
How Spermidine affects aging
Autophagy is the cell's recycling program: damaged mitochondria, misfolded proteins, and worn-out components get engulfed, broken down, and rebuilt into fresh parts. This housekeeping declines with age, and its failure is implicated in neurodegeneration, heart failure, and immune aging. Spermidine turns the program back on — it inhibits the acetyltransferase EP300, one of the main brakes on autophagy, mimicking at the molecular level what fasting does. This is why researchers class spermidine as a caloric-restriction mimetic: some of fasting's cellular signature, delivered through food.
The strongest preclinical result is cardiac. In the 2016 Nature Medicine study, adding spermidine to drinking water extended lifespan in mice and protected their aging hearts — less cardiac hypertrophy, better diastolic function, improved mitochondrial quality through enhanced mitophagy. Crucially, the benefits disappeared in animals whose autophagy machinery was disabled, pinning the mechanism to self-cleaning rather than some generic antioxidant effect. Spermidine has also extended lifespan in yeast, flies, and worms — an unusually consistent record across species.
The human evidence is observational and early. In the Bruneck cohort, higher dietary spermidine tracked with substantially lower mortality over 20 years — but people who eat wheat germ, legumes, and vegetables differ in many ways, so association is not proof. The one long randomized trial, SmartAge, gave older adults 0.9mg daily for a year and found no significant effect on its primary memory endpoint. Honest read: strong biology, encouraging epidemiology, and unproven supplementation — the food pattern is the defensible bet.
Dosage
RDA: No official RDA established. Typical diets provide on the order of 10mg per day, mostly from whole grains, legumes, vegetables, and aged or fermented foods.
Optimal for longevity: No target is established. In the Bruneck cohort, the top third of intake — roughly 12mg per day and up, from food — was associated with the lowest mortality. Supplement trials have used far smaller amounts (about 1mg daily as wheat-germ extract) with mixed results, so food-based intake is the better-supported route.
Food first, and it is genuinely easy: a spoonful of wheat germ on yogurt or oatmeal is the classic spermidine habit, and natto, mushrooms, aged cheese, legumes, and whole grains round out the list. These foods carry fiber, protein, and micronutrients along — the cohort-level benefits were seen at intakes achievable from diet alone, roughly 10-12mg or more per day.
Supplements are typically wheat-germ extracts delivering about 1-6mg. They appear safe at those doses, but the one year-long randomized trial at 0.9mg found no cognitive benefit, and doses matching the mouse studies have never been tested in humans. Your gut bacteria and your own cells also produce polyamines, so total exposure is more than diet alone. Treat capsules as unproven; treat the foods as unambiguously worth eating.

In the prospective Bruneck cohort, the top third of dietary spermidine intake had an all-cause mortality rate of 15.1 deaths per 1,000 person-years versus 40.5 in the bottom third; the adjusted hazard ratio was 0.76 per standard deviation of intake, a difference the authors likened to about 5.7 years of age. Observational — it cannot prove causation.
The honest counterweight: in 100 older adults with subjective cognitive decline, 0.9mg of wheat-germ-derived spermidine daily for 12 months produced no significant change in the primary memory outcome versus placebo. Exploratory signals on verbal memory and inflammation remain unconfirmed. Supplementation at this dose is not yet supported by trial evidence.
