Niacin (Vitamin B3)

The B vitamin your cells turn into NAD+.

Niacin is the dietary backbone of NAD+, the coenzyme every cell uses to turn food into energy and to power DNA repair and sirtuin activity. NAD+ levels decline with age, and that decline is tied to metabolic and mitochondrial dysfunction. A 2020 Cell Metabolism trial showed that plain niacin raised blood NAD+ up to eightfold and improved muscle strength in patients with systemic NAD+ deficiency — evidence that this cheap, decades-old vitamin is a genuine NAD+ booster.

How Niacin affects aging

NAD+ is the currency niacin buys. Through a short conversion pathway, nicotinic acid and nicotinamide become nicotinamide adenine dinucleotide — the electron carrier at the heart of energy metabolism. Every major route from food to ATP, from glycolysis to the citric acid cycle to oxidative phosphorylation in mitochondria, depends on NAD+ shuttling electrons. When NAD+ runs short, cells cannot extract energy from fuel efficiently, and mitochondria — the organelles whose decline is a core hallmark of aging — are the first to feel it.

NAD+ is not just a shuttle; it gets consumed. Sirtuins, the enzyme family linked to caloric restriction and longevity, burn NAD+ every time they regulate gene expression or repair metabolism. PARPs consume it to patch DNA damage, and the enzyme CD38 — which rises with age-related inflammation — degrades it outright. The result is a well-documented age-related fall in NAD+ availability, which is why NAD+ restoration has become one of the most active areas in aging biology, with niacin the oldest precursor of all.

The human evidence took a leap in 2020: in adults with adult-onset mitochondrial myopathy — a condition of systemic NAD+ deficiency — 750-1,000mg of niacin daily raised blood NAD+ up to eightfold, brought muscle NAD+ to control levels, increased muscle strength, and cut liver fat roughly in half. Two honest caveats: those patients were genuinely NAD+-deficient, and gram-level niacin is pharmacology, not nutrition — the large HPS2-THRIVE trial found high-dose niacin added side effects without cardiovascular benefit.

Dosage

RDA: 16mg NE for men, 14mg NE for women

Optimal for longevity: There is no established longevity dose. Most researchers focus on keeping NAD+ metabolism well supplied through food; the clinical trial that restored NAD+ used 750-1,000mg daily under medical supervision in patients with a diagnosed deficiency — a drug dose, not a dietary one.

Food first, and food is enough for baseline needs: a chicken breast alone covers most of the RDA, and your body also makes niacin from the amino acid tryptophan (about 60mg of tryptophan yields 1mg niacin equivalent). Meat, fish, peanuts, and whole or fortified grains make true deficiency rare in anyone eating adequate protein.

Forms matter above food doses. Nicotinic acid triggers the famous flush — harmless but unpleasant skin reddening — at doses as low as 50mg. Nicotinamide (niacinamide) does not flush and still feeds NAD+ synthesis, though it lacks nicotinic acid's lipid effects. Gram-level dosing of any form belongs under medical supervision: it can stress the liver and, per HPS2-THRIVE, adds risk without proven benefit in people who are not deficient.

The honest counterweight: in over 25,000 adults with vascular disease, high-dose niacin improved cholesterol numbers but did not reduce major vascular events — and increased serious adverse events including diabetes complications, infections, and bleeding. What is the difference between niacin and niacinamide?

Is the niacin flush dangerous?

Does niacin boost NAD+ like NR or NMN?

Should I take high-dose niacin for cholesterol or longevity?

Can I get enough niacin from food alone?
